Peptide synthesis produces a molecule with an intended sequence. Peptide sequencing investigates the order of its amino acids. Production, identification and purity assessment answer different questions.

What is peptide synthesis?

Chemical peptide synthesis assembles a chosen amino-acid sequence. In solid-phase synthesis, a growing chain is attached to a support during repeated chemical steps. The intended sequence is a design specification; it is not, by itself, an analytical result for the finished material.

What is peptide sequencing?

Peptide sequencing determines or infers the order of amino-acid residues. Tandem mass spectrometry examines fragment ions to obtain sequence information. Interpretation depends on the observed fragments and the method used; incomplete fragmentation can leave gaps.

A study combining collision-induced dissociation with electron transfer dissociation demonstrated how complementary fragmentation can improve sequence coverage. A mass measurement alone should not be presented as a complete sequence determination.

What should a research-material record explain?

Keep the stated compound identity, intended sequence and actual test results separate. When a report is available, check its sample identifier, laboratory, date, method and reported result. Ask what was tested rather than assuming that every possible quality attribute was measured.
