Retatrutide research: trials, outcomes and limitations

Retatrutide is studied as a GIP, GLP-1 and glucagon receptor agonist. Its publications address different questions about body weight and metabolic health. This guide covers selected studies, not a systematic review or a treatment guide.

What did the phase 2 obesity trial measure?

The 2023 randomised trial enrolled 338 adults and assessed changes in body weight over 48 weeks. It reported greater mean weight reduction in Retatrutide groups than with placebo. Gastrointestinal adverse events were common and dose-related; dose-dependent heart-rate increases were also reported.

An obesity-treatment trial must be read alongside its participant criteria, adverse events and follow-up duration. Weight loss measured in that setting is not a promised result for another material, and body weight is not the same as directly measured fat mass.

How is liver-fat research different?

A 2024 phase 2a report examined liver fat using MRI in participants with metabolic dysfunction-associated steatotic liver disease. An imaging change is a specific endpoint. It does not, on its own, demonstrate fewer long-term liver complications.

What does the phase 3 diabetes publication add?

The 2026 TRANSCEND-T2D-1 publication reports a randomised phase 3 study in people with type 2 diabetes. Its principal question concerns glycaemic control, including HbA1c, rather than the same population and endpoint as the earlier obesity trial. Comparisons between separate trials require attention to those differences.
